Further information: The plasmid was constructed by PCR amplifying the Y. enterocolitica sycD gene, flanked by 250 bp on both sides, and cloning it as a BamHI/HindIII fragment into the BamHI/HindIII opened pBC18R vector, resulting in pPW64. This plasmid was linearised at the unique NcoI site of sycD and subjected to exonuclease III digestion, resulting in pPW64Δ16, which carries an out-of-frame intragenic deletion, leading to premature termination at codon 44. This mutant allele was subcloned in pKNG101 and exchanged with the wild type allele of pYV227, resulting in pPW2269.
