|Further information:||This plasmid was derived from pPNT digested with Eco47III and HindII, followed by blunt-blunt ligation; in this way, the herpes simplex virus thymidine kinase gene was deleted.
This plasmid can be used for expression of heterologous genes under the control of the mouse PGK1 promoter by cloning them into the unique BstEII site. pPN1 can also be used as a negative control (e.g. for mouse E-cadherin, in combination with plasmid pPNEcad).
The neomycin resistance gene contains some modifications at the start of the coding sequence as compared to the wild type gene.
Since the plasmid contains the mouse PGK1 promoter and polyA sequence twice, one should be aware of the possibility of deletions due to homologous recombination.
When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide.
