Cloned DNA: Mouse interleukin 4 cDNA (IL4); mature sequence
Promoter: Phage λ major leftward promoter (λ PL)
Ribosome
binding site:
Ribosome binding site (RBS) of the Escherichia coli tryptophan operon attenuator peptide (trp)
Terminator: -
Selection marker: Ampicillin (amp)
Replicon: Escherichia coli plasmid pMB1 origin
Host range: Escherichia coli; use strains with a cI function, cIts for PL controlled expression
Parental clone: pPLc321; pMcMIL4m
Further information: The plasmid was constructed as follows: the ApaI (exonuclease-blunted)-BamHI fragment of pMcMIL4m, containing the mature mouse IL4 gene, was inserted into the NcoI (filled-in) and BamHI sites of pPLc321.
pPLcMIL4 is an expression plasmid, designed for expression of mature mouse IL4.
The nucleotide sequence of the mouse IL4 corresponds with the EMBL Nucleotide Sequence Database accession number M25892.1, except for 2 nucleotides (amino acid 56 of the mature sequence is CGT instead ofCGC and there is 1 T extra in the 3' UTR). The sequence matches with the one published in Lee et al.
The nucleotide sequence of the mouse IL4 3' UTR region has been sequenced (non-coding strand of pMcMIL4m from nucleotide position 4232 to 4358 in the orientation EcoRI-ScaI).
Other name of the plasmid is pPLctrpMIL4.
