Cloned DNA: Human interleukin 2 cDNA (IL2); mature sequence
Promoter: Phage λ major leftward promoter (λ PL)
binding site:
Ribosome binding site (RBS) of the phage Mu ner protein gene
Terminator: -
Selection marker: Ampicillin (amp)
Replicon: Escherichia coli plasmid pMB1 origin
Host range: Escherichia coli; use strains with a cI function
Parental clone: pAT153HIL2B; pGB299
Further information: pPLc436HIL2 is an expression plasmid, designed for expression of mature human IL2.
There is a correct fusion between the mature human IL2 sequence and the ATG codon and the phage Mu ner protein gene Shine-Dalgarno region supplied by pGB299.
The StuI site after the human IL2 gene (base position 496) can be used as a unique site; the second StuI site (base position 4818) cuts only in dcm-negative strains.
The nucleotide sequence of the human IL2 corresponds with the EMBL Nucleotide Sequence Database accession number X01586.1, except for the first codon of the mature sequence (GCA) that is GCG.
The nucleotide sequence of the phage Mu ner protein 5' UTR corresponds with the EMBL Nucleotide Sequence Database accession number X01586.1.
Other names of the plasmid are pPLcMuHIL201 and pPLc436MUHIL201.
