Genecorner:

-

Cloned DNA: -
Promoter: Phage λ major leftward promoter (λ PL)
Ribosome
binding site:
-
Terminator: Phage fd terminator
Selection marker: Ampicillin (amp)
Replicon: Escherichia coli plasmid pMB1 origin
Phage f1 origin
Host range: Escherichia coli; use strains with a cI function, cIts for PL controlled expression
Parental clone: pPLc2820; pSE380; pMc58
Further information: The plasmid was constructed by ligating four fragments: (i) the XbaI-AlwNI fragment of pMc58, containing a tandem terminator sequence of phage fd, the origin of replication of phage f1 as well as the Escherichia coli plasmid pMB1 replication origin, (ii) the AlwNI-EcoRI fragment of pPLc2820, containing the ampicillin resistance gene and the phage λ major leftward promoter (λ PL), (iii) the EcoRI-HindIII fragment of the superlinker of pSE380 and (iv) a XbaI-compatible/HindIII linker.
This plasmid is designed for λ PL promoter-regulated expression of fragments containing a ribosome binding site functional in E. coli.
The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727368.1.
