|Further information:||The plasmid was constructed as follows : pSP64MxpA was digested with XbaI / BglI and with BamHI / BglI to isolate the Mx-promoter and the SV40 intron and the polyA-sequences respectively. The human serotonin 5HT2 receptor was isolated as a NheI/BamHI fragment from pUCh5HT2R. Ligation of the three fragments resulted in pMxh5HT2R. This plasmid allows functional expression of the human serotonin 5HT2 receptor in L929s(A). The Mx promoter is inducible with IFNα and IFNβ.
When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide.
