Further information: The plasmid was constructed by inserting the BamHI/XhoI(filled in) fragment of pBLHIL6CAT, containing the human interleukin 6 promoter (hIL6), between the BamHI and HindIII (filled in) sites of pMa58.
The phasmid provides resistance to ampicillin, but is sensitive to chloramphenicol.
pMahIL6P contains the replication origin of the single-stranded DNA phage f1, so that it can be rapidly switched between the plasmid and the phage mode (ssDNA) of replication. The latter requires infection with a helper phage, e.g. M13KO7.
After mutagenesis, use mutS strains for primary transformation (e.g. sup(-) strain WK6mutS, sup(+) strain BMH71-18mutS); for segregation of possible mutants: sup(-) strains (e.g. WK6).
The nucleotide sequence of the genomic hIL6 DNA corresponds with the EMBL Nucleotide Sequence Database accession number AC073072.11. However, the C nucleotide at promoter position -174 (polymorphism mutant) was changed into a G nucleotide (wild type).
Other name of the plasmid is pMasHIL6.
