Further information: The Staphylococcus aureus spa gene coding for protein A was truncated 5' and 3', resulting in a fragment starting at nucleotide 175 and ending at nucleotide 915 of the spa coding sequence. This fragment, together with the polylinker (nucleotide position 4506 to 4557), was derived from pRIT21 and inserted into the unique BamHI and PstI site (nucleotide position 3776) of pMa58.
The phasmid provides resistance to ampicillin but is sensitive to chloramphenicol.
After mutagenesis, use mutS strains for primary transformation (e.g. sup(-) strain WK6mutS, sup(+) strain BMH71-18mutS); for segregation of possible mutants: sup(-) strains (e.g. WK6).
The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727587.1.
