|Further information:||The plasmid was constructed by isolating a 1.1 kb EcoRI fragment from placIq, carrying the lacIq gene and its promoter. The restriction ends were filled in using T4 DNA polymerase and HindIII linkers were ligated to the resulting blunt ends. The lacIq fragment was subsequently inserted into the HindIII-opened pMMB6 vector to give a self-repressible vector.
pMMB22 is a broad host range expression vector controlled by the tac promoter.
It can be used in all gram-negative bacterial species that support replication of the natural plasmid RSF1010.
