Further information: The plasmid was constructed by PCR amplifying the mCherry CDS from pRVCHYC-5. The last 30 bp of the Y. enterocolitica yscC gene and the start of the yscD gene were PCR amplified from pYV227. Both PCR fragments were joined by overlapping PCR. At the end of the Y. enterocolitica yscC CDS, the termination codon was deleted and a glycine/alanine linker (GGAGGAGG) and an XhoI restriction site were introduced, followed by the mCherry CDS. At the 3' end of the mCherry CDS, the last 10 codons of Y. enterocolitica yscC, including the termination codon, were introduced. The combined PCR products were cloned into the SalI/XbaI opened pKNG101 vector.
This is a mutator plasmid to introduce yscC-mCerry.
Other name of the plasmid is pKNG101-mCherry-yscC.
