|Further information:||The human immunoglobulin γ1 gene encoding the constant domains of the heavy chain (hIG1) was isolated from the partial HaeIII-AluI human genomic library (Maniatis) and subsequently cloned as a HindIII-PvuII fragment of approx. 2770 bp in the HindIII-PvuII opened pBR322 vector.
The 5' flanking region (5FR) is preceding the CH1 exon in the genomic DNA.
pBRHIG1 was designed to add V-domains to the constant portion in an eukaryotic vector, so that it becomes useful in eukaryotic cells.
The nucleotide sequence of hIG1 was taken from Ellison et al. (1982).
Since part of the nucleotide sequence of the 3' flanking region of the hIG1 gene (from nucleotide position 2038 to approx. 2802 in pBRHIG1) is unknown, there is uncertainty as to the cutting frequency of the restriction enzymes indicated.
Other name of the plasmid is pBRHuIG1.
